Effect of Aloe vera extract on the improvement of the respiratory activity of leukocytes of matrinxã during the transport stress R. Bras. Zootec.
Zanuzzo,Fábio Sabbadin; Biller-Takahashi,Jaqueline Dalbello; Urbinati,Elisabeth Criscuolo.
This study evaluated the effect of extract of Aloe vera in the transport water of matrinxã (Brycon amazonicus) fish on stress response and leukocyte respiratory activity. Fish was transported for 4 h in water containing Aloe at levels 0; 0.02; 0.2 and 2 mg/L, and sampled before transport 2, 4, 24 and 96 h after for determination of plasma glucose and respiratory activity of leukocytes. An additional in vitro assay was conducted with another fish species, pacu (Piaractus mesopotamicus), to test the respiratory burst of leukocytes exposed to Aloe extract (0.0, phosphate-buffered saline (PBS) only) at 0.1, 0.2, 0.5 and 1 mg/L). Plasma glucose increased after 2 and 4 h of transport and returned to control levels within 24 h, but the addition of Aloe in the...

Evidence in oyster of a plasma extracellular superoxide dismutase which binds LPS ArchiMer
Gonzalez, Marcelo; Romestand, Bernard; Fievet, Julie; Huvet, Arnaud; Lebart, M; Gueguen, Yannick; Bachere, Evelyne.
We have characterized in the oyster Crassostrea gigas an extracellular superoxide dismutase (Cg-EcSOD) which appears to bind lipopolysaccharides (LPS). The protein has been purified from the oyster plasma and identified as a Cu/ZnSOD according to its N-terminal sequencing and biological activity. Cg-EcSOD expression and synthesis are restricted to hemocytes as revealed by in situ hybridization and immunocytochemistry. Cg-EcSOD-expressing hemocytes were seen in blood circulation, in connective tissues, and closely associated to endothelium blood vessels. Cg-EcSOD presents in its amino acid sequence a LPS-binding motif found in the endotoxin receptor CD14 and we show that the protein displays an affinity to Escherichia coli bacteria and with LPS and Lipid A....

Modulation des paramètres hémocytaires par la nutrition de l'huitre creuse Crassostrea gigas. Implication dans les mortalités estivales ArchiMer
Delaporte, Maryse.
In the context of Pacific oyster C. gigas summer mortality, the present study demonstrated that the reproductive effort reduced not only the energetic parameters (carbohydrates and CEA) but also the immune parameters (concentration, phagocytosis and adhesive capacity of hemocytes). Increasing food supply from 4% to 12% (of oyster dry weight) did not counteract this phenomena as most of the additional energy was dedicated to the gametogenesis processes. However, after the gametogenesis, the 12% diet clearly favored energy storage restoration and hemocyte production. Hemocyte parameters were also modulated by the food quality. Indeed, hemocyte concentration tended to increase 20:5n-3 and/or 20:4n-6 were supplied by micro-algae or artificial diets. When these...

Use of OmpU porins for attachment and invasion of Crassostrea gigas immune cells by the oyster pathogen Vibrio splendidus ArchiMer
Duperthuy, Marylise; Schmitt, Paulina; Garzon, Edwin; Caro, Audrey; Rosa, Rafael D.; Le Roux, Frederique; Lautredou-audouy, Nicole; Got, Patrice; Romestand, Bernard; De Lorgeril, Julien; Kieffer-jaquinod, Sylvie; Bachere, Evelyne; Destoumieux-garzon, Delphine.
OmpU porins are increasingly recognized as key determinants of pathogenic host Vibrio interactions. Although mechanisms remain incompletely understood, various species, including the human pathogen Vibrio cholera, require OmpU for host colonization and virulence. We have shown previously that OmpU is essential for virulence in the oyster pathogen Vibrio splendidus LGP32. Here, we showed that V. splendidus LGP32 invades the oyster immune cells, the hemocytes, through subversion of host-cell actin cytoskeleton. In this process, OmpU serves as an adhesin/invasin required for beta-integrin recognition and host cell invasion. Furthermore, the major protein of oyster plasma, the extracellular superoxide dismutase Cg-EcSOD, is used as an opsonin mediating the...

WBC count and functional changes induced by co-administration of clofazimine and clarithromycin, in single and multiple doses, in Wistar rats BJPS
Paina,Flávia Aparecida; Miranda,Julise Cunha; Tavares,Cristiane Fernandes de Freitas; Queiroz,Regina Helena da Costa; Souza,Ana Maria de.
Clofazimine and clarithromycin are used to treat leprosy and infections caused by Mycobacterium avium complex. Little data on the toxicity of co-administration of these two drugs are available. Here we evaluated the potential adverse effects of polytherapy with these two drugs in male Wistar rats by determining WBCs counts and other blood cell counts, neutrophilic phagocytosis, and burst oxidative, by flow cytometry. We observed an increase in WBCs, in multiple-dose regimens, and in polymorphonuclear cells, in both single- clarithromycin only and multiple dose regimens. We also observed a reduction in mononuclear cell counts in single and multiple doses. The drugs seem to reverse the mononuclear and polymorphonuclear cell ratio. An increase in oxidative...
